Decoding Kupffer Cell Heterogeneity in HBV-Associated Hepatocellular Carcinoma

This study leveraged single-cell RNA sequencing to characterize Kupffer cell subtypes in HBV-related hepatocellular carcinoma. Using Seurat and CellChat, we performed clustering, cell–cell interaction mapping, and functional enrichment to uncover key ligand–receptor pathways driving tumor progression.

Objective: To map the heterogeneity and communication networks of Kupffer cells within the HBV-HCC tumor microenvironment
Key Results:
CellChat networks in HCC confirmed the Kupffer-endothelial-stellate axis as a fundamental component of TME remodelling. This study presents the first comprehensive communication map of Kupffer cell subtypes in HCC, emphasising new immunometabolic and angiogenic pathways with therapeutic relevance. Our findings reveal significant flaws in tumour-associated macrophages and provide a foundation for developing liver cancer therapies that target macrophages.
